Checking transmission oil level on my 1974 DT360 was created by Roland Wheeler

Hi Yamaha Enduros,
The bike is running awesome thanks to all of your help. I do have a question, and I cannot seem to locate a previous answer (probably not putting in the proper search criteria). Do I check the level with the dipstick screwed in, or just inserted? 
Thank you, and sorry for the ( maybe) silly question that I should know, but my manual does not specify.

Just dip it with bike vertical, cheers. ps, if its cold & oil thicker it might take a minute to settle if you stand it vertical after sitting on the side stand all night--call it kick stand in US.
